Choose a preservation method by working backwards from the sample, required storage time and downstream use. Short-term fresh or hypothermic preservation, cryopreservation and fixation solve different problems. The correct method is the one validated to retain the quality attributes your assay needs.
There is no universal ‘best’ preservation method
A tissue biopsy intended for viable cell isolation should not automatically be handled like a sample intended only for fixed histology. An assay-ready plate should not be treated like a long-term cell bank. A living organoid needed next week has different requirements from a master stock intended for years of storage.
Preservation choices become clearer when five questions are answered in order.
Five questions to ask before choosing
- What is the biological material? Whole tissue, dissociated cells, blood, leukapheresis, organoids and plate-based models respond differently.
- What must be retained? Viability alone may be insufficient. Consider phenotype, architecture, marker expression, molecular integrity and function.
- How long is preservation required? Hours, days, weeks and years point towards different approaches.
- In what format must the sample arrive? Vials, plates, inserts, bags and intact tissue pieces create different handling and recovery needs.
- What logistics can the sending and receiving sites reliably support? Equipment, customs routes, weekend working and operator expertise all matter.
Match the method to the intended use
For immediate or short-term use, fresh transport in a validated medium and temperature range may be appropriate. Non-cryogenic hypothermic preservation can extend this window while keeping the sample in a recoverable state. Cryopreservation is valuable for long-term banking and many established cell workflows, but it introduces freezing, thawing and post-thaw recovery steps. Fixation is suitable when preserved morphology or molecular targets are required, and live recovery is not.
These approaches are not rivals in every situation. A robust organisation may use several of them at different stages of the same programme.

A practical selection rule
Use the least disruptive method that reliably meets the required storage window and endpoint. Avoid adding a freeze-thaw cycle to a short journey if it creates unnecessary recovery work. Equally, do not force a short-term preservation system into a long-term banking role for which it was not designed.
Before implementation, compare the proposed method with the current control under a representative journey. Include the end-of-window timepoint, a realistic delay and the actual receiving-site process. Preservation should make the workflow more controlled, not merely move complexity from one laboratory to another.
