Title

Includes non-crosslinked gels, gelation buffers and dissolution buffer for the encapsulation, storage and release of cells in 48-well plate format. Modified Protocols Air-sensitive cells are cell types that are compromised when exposed to the air during medium change (eg. some neuronal cultures). Cardiomyocytes have extra sensitivity to ion flux. The WellReady protocol has been amended to suit these particular cell types.

QUICK AND EASY TO USE

With cells, organoids and tissue models in place, three gel layers are added and cured using a Gelation Buffer. These gel layers offer physiochemical and mechanical protection during hypothermic storage and shipment. When plates are ready to be used, a gentle Dissolution Buffer is added to each well to dissolve the gels. The plates are returned to culture conditions and once recovered they are ready to be used for downstream assays or continued culture.

APPLICATIONS

Preserve complex multicellular model viability, phenotype and function 

Liver organoids preserved at 20°C for 5 days using WellReady™. Following preservation, the organoids were released from WellReady™ and returned to culture for 72 hours before carrying out assays. Left - Cell viability, Middle -  functional Cytochrome P450 1A2 activity right: Organoids were returned to culture for overnight, fixed, and stained for the hepatocyte cholangiocyte marker - Cytokeratin-19 (CK19, green) and Hoechst nuclear stain. 

 

Protect against mechanical/shearing forces during shipment 

iPSC-derived cortical neurons were preserved using WellReady™ for 3 days and return-shipped by courier in CRT packaging at 15-20°C. Cells were retured to normal culture conditions and live/dead stained - Calcein-AM (live indicator; green) and Ethidium Homodimer-1 (dead indicator; red).  

 

Retain drug screening sensitivity across multiple assays 

Left - HEK-293 cells were stored and shipped using WellReady™ for 5 days at room temperature. Following release, cells were incubated for 24 hours before examining calcium flux in response to ATP using as fluo-4 assay kit. Middle - HEK-293 cells transiently transfected with a DDR1 kinase Nanoluc Fusion element before storage for 5 days at room temperature. DDR1-NanoLuc in the presence of a substrate gives a luminescent signal (BRET), dasatinib completes with the substrate binding site, increasing concentrations of which reduces luminescent signal. Right - HEK-293 cells transiently transfected with a cyclic AMP-response element – firefly luciferase cDNA construct were stored and shipped for 5 days at room temperature before assessing cAMP levels in response to Forskolin treatment.
